Doctors Best SAM e

S-Adenosyl Methionine (SAM-e) is a unique molecule that is completely utilized by the body. Derived from the amino acid methionine, it is one of the most important methyl donors in the central nervous system and the liver. Methylation (the donation of methyl groups) is an essential process that occurs billions of times daily and facilitates ideal function of a number of organ systems. Involved in numerous biochemical reactions in tissues including those of the liver, joints, and brain, SAM-e has also been found to decrease in tissue with age. Required for the biosynthesis of critical neurotransmitters and hormones, SAM-e catalyzes the production of epinephrine, melatonin, glutathione, and the amino acids cysteine and taurine through its methyl donation. SAM-e is also known to upregulate the production of critical membrane phospholipids including phosphatidylcholine. As a sulfur donor, SAM-e is critical for the formation of proteoglycans in joint connective tissue.

Best Supplement design effects potency during the manufacture of SAM-e, two forms are produced known as the S,S and the R,S isomers. The S,S isomer is the biologically active form in the human body, whereas the R,S form is considered to be inactive. Doctor’s Best SAM-e utilizes Italian SAM-e to yield the highest available percentage of the active pure S,S form on the market and contains an average of 74% of the active S,S form. Other SAM-e products contain as little as 50% of the active form. A higher content of the active form yields a more potent product.

The SAM-e molecule is highly unstable and will degrade quickly in conditions of heat, light, and moisture. To increase its stability, it must be formed into a salt. It is important to note that many different types of SAM-e salt forms are found in dietary supplements. Doctor’s Best SAM-e features the form most extensively used in clinical trials—the tosylate disulfate salt form of SAM-e. To get to the intestine (where it is best absorbed), an intact capsule of SAM-e must pass through the acidic environment of the stomach. Enteric-coated capsules are used to protect SAM-e from the damaging effects of stomach acid. Doctor’s Best SAM-e tablets are enteric-coated to maximize the absorption—and thus the potency—of the SAM-e.

Enhances Mood

Over the past three decades, SAM-e has been studied for its great potential in enhancing mood. In randomized controlled trials SAMe appears to work as well as the frequently prescribed tricyclic antidepressants (imipramine, amitriptyline, desipramine) in easing depression. Just as importantly, it works faster, often starting to improve mood within a week. This contrasts significantly with standard antidepressants, whose effects can take several weeks to become apparent. In a University of California study of 17 severely depressed adults, 62% of the participants who took SAMe for four weeks (1,600 mg daily) showed significant improvements in symptoms, compared to 50% of those who used desipramine (a conventional Antidepressant). Unlike many prescription antidepressants, which often cause unpleasant side effects such as as drowsiness, dry mouth, and constipation, SAMe appears to cause few if any side effects. SAMe may also be a good choice for women who take oral contraceptives and would like to try a herbal supplement to treat depression. SAMe has not been demonstrated to adversely effect the efficacy of oral contraceptives, as has St. John’s Wort.

Promotes Joint Comfort and Mobility

As a sulfur donor to connective tissue, SAM-e plays a major role in maintaining the integrity of cartilage tissue. The specialized cells that make up cartilage (chondrocytes) are bound together by a matrix of collagen fibers and proteoglycans. An in vitro experiment assessing the actions of SAM-e in cultured human articular chondrocytes demonstrated sulfate residue incorporation into the chondrocytes, as well as proteoglycan synthesis 60% higher than control levels. Based on these observations, the researchers concluded that SAM-e promotes the growth and health of cartilaginous connective tissue. While in vitro experiments are useful, evidence of benefit in the human body is usually best shown through clinical research. In a large double-blind trial examining 734 individuals, SAM-e was given orally at a dose of 1200 mg daily for 30 days. SAM-e was shown to significantly promote joint comfort compared to placebo, with a high level of tolerability and low incidence of side effects. In a long-term trial lasting 24 months, 600 mg per day of SAM-e was orally administered to 108 participants for the first two weeks, followed by 400 mg daily for the remainder of the trial (97 participants completed the full 2 years of supplementation). Significant and relatively rapid enhancements in joint comfort were noted, with excellent long-term tolerance and continued benefits seen throughout the 2 years.

Phosphatidylcholine is an important component for the normal function of our cell membranes. It is formed through the methylation of phosphatidyl ethanolamine (a phospholipid), with the help of SAM-e. Increased levels of SAMe and phosphatidylcholine are associated with more “fluid” cell membranes, facilitating the movement of proteins (including receptors) within the lipid bilayer of cell membranes, providing for better cell signaling. This may, in part, explain how SAM-e is associated with increased proteoglycan synthesis—and therefore with joint comfort and mobility.

Supports Liver Health and Detoxification

It is found SAM-e supplementation can profoundly influence liver function due to its role as the major methyl donor in the liver, as well as its lipotropic, or fat-removing, activity. SAM-e also helps the liver to confront oxidative stress by enhancing the production of glutathione. A number of trials have been conducted to demonstrate the ability of SAM-e to support liver detoxification functions towards enhancing liver health; one means to this end is the ability of SAM-e to rapidly increase the activity of enzymes needed to support liver detoxification. Studies indicate that supplementing with SAM-e improves liver-function tests and prevents or reverses liver toxicity induced by drugs, alcohol and various chemicals.
